The nucleophosmin (NPM1) mutation (a four base pair insertion in exon 12) is found in half of AML cases that have a normal karyotype; its presence is associated with a good prognosis when FLT3 ITD is absent, and an intermediate prognosis if FLT3 ITD is present.
